Background: Many previous studies have shown that soybean WRKY transcription factors are involved in the plant response to biotic and abiotic stresses. Phakopsora pachyrhizi is the causal agent of Asian Soybean Rust, one of the most important soybean diseases. There are evidences that WRKYs are involved in the resistance of some soybean genotypes against that fungus. The number of WRKY genes already annotated in soybean genome was underrepresented. In the present study, a genome-wide annotation of the soybean WRKY family was carried out and members involved in the response to P. pachyrhizi were identified. Results: As a result of a soybean genomic databases search, 182 WRKY-encoding genes were annotated and 33 putative pseudogenes identified. Genes involved in the response to P. pachyrhizi infection were identified using superSAGE, RNA-Seq of microdissected lesions and microarray experiments. Seventy-five genes were differentially expressed during fungal infection. The expression of eight WRKY genes was validated by RT-qPCR. The expression of these genes in a resistant genotype was earlier and/or stronger compared with a susceptible genotype in response to P. pachyrhizi infection. Soybean somatic embryos were transformed in order to overexpress or silence WRKY genes. Embryos overexpressing a WRKY gene were obtained, but they were unable to convert into plants. When infected with P. pachyrhizi, the leaves of the silenced transgenic line showed a higher number of lesions than the wild-type plants. Conclusions: The present study reports a genome-wide annotation of soybean WRKY family. The participation of some members in response to P. pachyrhizi infection was demonstrated. The results contribute to the elucidation of gene function and suggest the manipulation of WRKYs as a strategy to increase fungal resistance in soybean plants. No Centro-Oeste e Norte, onde é comum a ocorrência de elevadas taxas de precipitação pluvial, altas temperaturas e manutenção de solo e ar saturados por períodos prolongados, o processo de infecção-doença-disseminação da gomose é acelerado, causando elevada mortalidade de plantas mais precocemente. Assim, o trabalho foi realizado com o objetivo de avaliar variáveis de crescimento vegetativo de limeira ácida clone ?Tahiti CNPMF 02? enxertada em 12 porta-enxertos. Os porta-enxertos foram gerados e pré-selecionados pelo Programa de Melhoramento Genético de Citros da Embrapa e avaliados no município de Sorriso, MT. Os porta-enxertos utilizados foram: citrandarins ?Indio? [C. sunki (Hayata) hort ex Tanaka x Poncirus trifoliata (L.) Raf. ?English?] (CTRI) e ?San Diego? (C. sunki x P. trifoliata ?Swingle?) (CTRSD), citrumelo ?Swingle? (C. paradisi Macfad. x P. trifoliata) (CTSW), limoeiro ?Cravo?, clones ?Santa Cruz? e ?CNPMF-003?, tangerineira ?Sunki Tropical? (C. sunki) e os híbridos HTR - 069, TSKC x (LCR x TR) - 059, LVK x LCR - 038, TSKC x TRFD 003, TSKC x CTSW ? 028 e 041. O delineamento experimental foi de blocos inteiramente casualizados, quatro repetições e doze tratamentos. As parcelas foram constituídas por cinco plantas. Os dados foram submetidos à ANOVA e as médias ao teste de Scott-Knott à 5% de significância. Com quatro anos e seis meses de idade foram avaliados os seguintes caracteres: altura de planta, diâmetro do caule (10 cm acima e abaixo da linha de enxertia), diâmetro e volume da copa, este calculado pela fórmula V=2/3 x [(π x D/4) x H], o Índice de Vigor Vegetativo (IVV) foi calculado pela fórmula IVV = [(H x 100) + (D x 100) + (DPE)]/100. No conjunto das características avaliadas, observou-se que os limoeiros ?Cravo Santa Cruz? e o ?Cravo CNPMF-003?, TSKC x CTSW ? 028, o citrumelo ?Swingle? e os citrandarins ?San Diego? e ?Índio? foram significativamente superiores à tangerineira ?Sunki Tropical? e aos híbridos TSKC x (LCR x TR) ? 059, LVK x LCR ? 038, TSKC x TRFD 003, HTR ? 069 e TSKC x CTSW - 041, alcançando uma altura média de 3,62 m e diâmetro de 12,59 cm para o porta-enxerto limoeiro ?Cravo Santa Cruz?, enquanto que o crescimento foi menor para os demais porta-enxertos, com altura média de plantas, de 3,17 m e diâmetro do enxerto de 11,62 cm para o híbrido TSKC x (LCR x TR) ? 059 de menor crescimento vegetativo.
